Some of the more popular reasons people seem to use CBD is for sleep, anxiety, and pain. In this mini-series, we will discuss each of these topics, in regard to how terpenes work alongside CBD to help in each area. The combination of these terpenes and cannabinoids is what’s known as the Entourage Effect. In short, the National Institutes of Health (NIH)  explains, “In 1998, Professors Raphael Mechoulam and Shimon Ben-Shabat posited that the endocannabinoid system demonstrated an “entourage effect” in which a variety of “inactive” metabolites and closely related molecules markedly increased the activity of the primary endogenous cannabinoids, anandamide and 2-arachidonoylglycerol.”

For starters, the NIH just announced a few months ago that $3 million of funding has been awarded to the National Center for Complementary and Integrative Health (NCCIH), which is part of the NIH, to research the potential role of cannabinoids and terpenes in pain management because, “there’s an urgent need for more effective and safer options” than the current methods of opioids.

Myrcene:

A study by the NIH states that, “Molecular docking studies suggest a non-covalent interaction site for Myrcene in TRPV1 and identifies key residues that form partially overlapping Myrcene and Cannabidiol binding sites. We identify several non-Cannabis plant-derived sources of Myrcene and other compounds targeting nociceptive TRPs using a data mining approach focused on analgesics suggested by non-Western Traditional Medical Systems. These data establish TRPV1 as a target of Myrcene and suggest the therapeutic potential of analgesic formulations containing Myrcene.” In other words, not only does it suggest that Myrcene may be a therapeutic option for pain, but also says that is shares binding sites with CBD.

β-Caryophyllene (β-CARY):

This study by the NIH says: “β-CARY is known to be the main constituent of Cannabis sativa essential oil [231] as well as to possess antiarthritic effect [232] and noteworthy anti-inflammatory activity against carrageenan- and PGE1-induced edema in rats [233]. With this background, [234] examined the contribution of opioid and peripheral cannabinoid (CB) systems in the antinociceptive action produced by β-CARY as well as its action in combination with the opioid agonist morphine.” It then continues on to say, “Finally, it was also observed a synergistic antinociceptive interaction between β-CARY and morphine, fact that may be an interesting therapeutic alternative to minimize risk of undesirable side-effects caused by this opioid analgesic.”

Linalool:

This study states some intriguing evidence. “The results show that this compound induced a significant reduction of the acid-induced writhing at doses ranging from 25 to 75 mg/kg. Such effect was completely reversed both by the opioid receptor antagonist naloxone and by the unselective muscarinic receptor antagonist atropine. In the hot plate test, only the dose of 100 mg/kg of (-)-linalool resulted in a significant effect. (-)-Linalool induced a dose dependent increase of motility effects, thus ruling out the confounding influence of a possible sedative effect. The more pronounced effect of (-)-linalool on the writhing test with respect to the hot plate test is consistent with the observation that (-)-linalool possesses anti-inflammatory activity. Finally, the activation of opioidergic and cholinergic systems appears to play a crucial role in (-)-linalool-induced antinociception.”

It is important to remember that these dosages used in the studies are not combined with other terpenes and cannabinoids, which, by other research, is shown to enhance the effect of each other.
